Scooby-Doo
Scoobert "Scooby" Dooby Doo is a Great Dane who is a part of Mystery Inc. Unlike most dogs, Scooby has the unique ability of being able to talk. Some series show him speaking broken English and putting an "R" in front of every word (much like Astro in The Jetsons), while later series show him speaking perfect English. Scooby is Shaggy's dog, and lives with Shaggy in his house, except for in A Pup Named Scooby-Doo, where Scooby lives in a separate dog house that is much bigger on the inside than on the outside (sort of like the TARDIS from Doctor Who). Fred Silverman came up with Scooby's name in 1968, getting the idea from the syllables "doo-bee doo-bee doo" in the song "Strangers in the Night" by Frank Sinatra. Scooby's catchphrases are "Ruh-Roh!" and "Scooby Dooby Doo!"

Velma is the brains of the group, who often puts together the clues to uncover the solution to the mystery. Velma's catchphrase is "Jinkies." In early series, Velma is shown to be a bit of a know-it-all, but is toned down in most media beginning with The Scooby-Doo Show. In A Pup Named Scooby-Doo, Velma is characterized as being shy and quiet. In What's New Scooby-Doo?, Scooby-Doo! Mystery Incorporated and Be Cool Scooby-Doo, she has a dry and sardonic personality, while still retaining her high level of intelligence. In later media, Velma is shown to as having a rigid belief in scientific facts, sometimes to the point of discounting anything supernatural without an explanation based on logic.
In Scooby-Doo! Mystery Incorporated season 1, Velma and Shaggy begin dating. However, they break up after ten episodes, due to Velma being overly controlling. In season 2 of Scooby-Doo! Mystery Incorporated, Velma is implied to have a crush on Marcie, who is a recurring character whom Velma befriends. Tony Cervone confirmed in 2020 that the two were officially thought of as a couple by the writers, alluding to that being the explanation why Velma and Shaggy's relationship didn't work out. In Trick or Treat, Scooby-Doo!, Velma is shown as having a crush on Coco Diablo, a criminal mastermind. This is the first official piece of Scooby media characterizing her as being romantically interested in women. The series Velma also characterizes her as being bisexual.

Fred Jones
Frederick "Fred" Herman Jones is the unofficial leader of the gang who always wears a white shirt and orange ascot. Often, he is the one to set the traps for the monster. In the early series, Fred is characterized as being the stoic leader of the gang. Fred has a sassier side in some episodes, particularly in The New Scooby-Doo Movies. Frank Welker has voiced Fred in every series and movie (other than the theatrical movies and Velma), and is the only original Scooby-Doo voice actor remaining in the voice cast.
Starting in 1988, Fred's character was significantly rewritten to be sillier. In this series, Fred is a very zany child who believes in wild legends, most of them derived from a conspiracy theory newspaper, The National Exaggerator. A running gag in this series is that he suspects the neighborhood bully, Red Herring, for being the culprit to every mystery. In this series, he's depicted as being a trap-obsessed teenager who struggles with his emotions. In Be Cool, Scooby-Doo!, Fred is more of an authoritative leader who likes structure, and is at times a control freak. Most recent DTVs have characterized him as a friendly leader with a slightly awkward and goofy side to him.
Some adaptations show Fred as having a crush on Daphne Blake, but it is often just hinted at that they like each other. In Scooby-Doo Mystery Incorporated, he and Daphne are officially dating, and at one point become engaged. However, this nickname is only used when the gang is annoyed with her, and is phased out after the first few series (despite Daphne being "danger-prone" staying in public consciousness to this day). In the second season of The New Scooby-Doo Movies, her characterization briefly shifted to being more cowardly like Shaggy and Scooby; however, this was quickly dropped. Daphne is shown to be a bit vain and self-conscious about her looks through Scooby-Doo, Where Are You?, but this was also removed from her characterization after this series.

These character changes were likely due to the rise of the "action girl" trope which was popular at this time, with shows like Buffy the Vampire Slayer and Tomb Raider.

Vincent is a mystical warlock who meets the gang when their plane crashes in The 13 Ghosts of Scooby-Doo. After Shaggy and Scooby are tricked into opening the Chest of Demons, Vincent helps the gang capture the 13 ghosts. He often contacts the gang through a crystal ball. Vincent appears in every episode of 13 Ghosts.
However, in Scooby-Doo Mystery Incorporated, Van Ghoul is an actor in horror films instead of an actual warlock. He has acted in over 400 horror movies in this series, and many clips of his movies are shown throughout the series. In "Nightfright," Shaggy and Scooby win an essay contest and get to have dinner with Van Ghoul, but instead of having dinner, they end up having to capture "Nightfright," a demon who wants to kill Van Ghoul. In "Theater of Doom," the gang helps Vincent put on a play about Friar Serra and his donkey Porto, but the play ends up being spooked by the real Mummy of Friar Serra.

In Scooby-Doo and the Legend of the Vampire, they are a part of the Vampire Rock Music Festival, where the gang meets up with them again to solve the mystery of a group of vampires. They are kidnapped during the festival by a vampire rock band. The gang runs into them again at Fortescu Castle in the What's New, Scooby-Doo? episode "The Vampire Strikes Back," where their music video filming is ruined by a vampire.
In Scooby-Doo! Mystery Incorporated, Velma is an avid fan of The Hex Girls. In "In Fear of the Phantom," the gang attends a Hex Girls concert, and end up helping them solve the mystery of a phantom who tried to sabotage the concert. In this episode, Daphne briefly joins the band onstage under the persona "Crush," as bait for the phantom. The girls also help the gang solve the mystery of Rude Boy and Ska-Tastics in "Dance of the Undead" by defeating the ghosts of the Ska-Tastics. Later, the girls also help the gang find the key of water with the Planispheric Disk, by using their harmonizing voices to help hit the correct note to unlock the disk.
In Scooby-Doo and Guess Who?, the gang helps out Thorn, Luna and Dusk stop the ghost of Ester Moonkiller, who has put Thorn and Luna into a trance of being ultra girly and ungothy. Thorn is portrayed as much more violent in this adaption. She admits to trashing her own house eight times in the past week for fun, and has a tranquilizer gun for if she gets too wild, labeled "in case of Thorn."
